Mechanism of Action

Mazdutide activates two receptor pathways. GLP-1 receptor activation in the hypothalamus suppresses appetite signaling and slows gastric emptying to prolong post-meal satiety. Glucagon receptor activation in the liver stimulates glycogenolysis, increases resting energy expenditure through thermogenesis, and promotes hepatic fat oxidation via upregulation of CPT1a and other beta-oxidation enzymes. The combined effect reduces energy intake while increasing energy expenditure — a dual mechanism not achieved by GLP-1-only or GLP-1/GIP compounds.

FAQ

Frequently Asked Questions

What is mazdutide?
Mazdutide is a dual GLP-1/glucagon receptor agonist peptide. It activates both GLP-1 receptors (appetite suppression) and glucagon receptors (energy expenditure) simultaneously. It is being studied for metabolic regulation and body composition effects in research settings.
How is mazdutide different from semaglutide?
Semaglutide targets only GLP-1 receptors for appetite suppression. Mazdutide targets both GLP-1 and glucagon receptors, adding energy expenditure and hepatic fat oxidation effects that GLP-1-only agents do not provide.
How is mazdutide different from tirzepatide?
Tirzepatide pairs GLP-1 with GIP receptor activation, while mazdutide pairs GLP-1 with glucagon receptor activation. The glucagon component promotes thermogenesis and liver fat oxidation, while GIP enhances fat metabolism and insulin sensitivity. They are mechanistically distinct dual agonists.
What has mazdutide shown in published studies?
Published studies have demonstrated up to 18% body weight reduction over 48 weeks, along with reductions in hepatic fat content, improvements in lipid profiles, and improved glycemic control markers. The hepatic fat reduction is particularly notable.
How should mazdutide be stored?
Store lyophilized mazdutide at -20°C for long-term storage or 2-8°C for short-term use. Once reconstituted with bacteriostatic water, refrigerate and use within 28 days.
Is mazdutide available for research purchase?
Yes. Mazdutide is available as a research-use-only peptide. It is not approved for therapeutic use. All research should be conducted under appropriate protocols.
