Mechanism of Action

SS-31 is a cell-permeable peptide that rapidly crosses membranes without requiring active transport or mitochondrial membrane potential. Once inside mitochondria, it binds selectively to cardiolipin in the inner mitochondrial membrane through electrostatic and hydrophobic interactions between its alternating aromatic-cationic motif and cardiolipin's anionic headgroups. This binding stabilizes cardiolipin microdomains, prevents pathological cardiolipin peroxidation, maintains cytochrome c in its native electron carrier conformation, and optimizes electron transfer through complexes III and IV of the respiratory chain.

Storage & Handling

FAQ

Frequently Asked Questions

What is SS-31?
SS-31 (Elamipretide) is a synthetic tetrapeptide with the sequence D-Arg-Dmt-Lys-Phe-NH2. It is a mitochondria-targeted peptide that concentrates in the inner mitochondrial membrane where it binds cardiolipin, stabilizes cytochrome c, and optimizes electron transport chain function. It is one of the most researched mitochondrial peptides. For research use only.
How does SS-31 differ from other mitochondrial peptides?
While most mitochondria-targeted compounds accumulate in the matrix driven by membrane potential, SS-31 selectively binds cardiolipin in the inner mitochondrial membrane. This gives it a unique mechanism of action focused on stabilizing cristae structure and electron transport chain supercomplexes rather than acting as a matrix-localized antioxidant.
What has SS-31 been studied for?
SS-31 has been investigated in preclinical and clinical research for age-related mitochondrial dysfunction, heart failure with reduced ejection fraction, primary mitochondrial myopathy, Barth syndrome, renal ischemia-reperfusion injury, skeletal muscle aging, and age-related macular degeneration.
How should SS-31 be stored?
Store lyophilized SS-31 at -20 degrees C for long-term storage or 2-8 degrees C for short-term use. Once reconstituted, refrigerate and use within 28 days. Protect from light and avoid repeated freeze-thaw cycles.
