Mechanism of Action
KPV enters cells and inhibits NF-κB activation by preventing nuclear translocation of inflammatory transcription factors. This reduces downstream production of pro-inflammatory cytokines including TNF-α, IL-6, and IL-1β, attenuating the inflammatory cascade across multiple tissue types.
Overview
KPV is one of the smallest bioactive peptides studied in inflammatory research. This tripeptide, consisting of the amino acid sequence Lys-Pro-Val, is derived from the C-terminal region of alpha-melanocyte-stimulating hormone (alpha-MSH), a neuropeptide with well-documented anti-inflammatory activity. Preclinical research has demonstrated that KPV inhibits NF-κB signaling, a master regulator of the inflammatory response. Studies show it reduces the production of pro-inflammatory cytokines including TNF-α, IL-6, and IL-1β. Its compact size allows efficient cellular uptake and tissue penetration compared to larger peptide sequences. KPV has been studied extensively in models of gut inflammation, including IBD and colitis, where it has shown effects on mucosal healing and inflammatory marker reduction.
