Mechanism of Action

LL-37 disrupts microbial membranes through electrostatic interaction with negatively charged phospholipids. It also activates FPRL1 receptors on immune cells, promoting chemotaxis and cytokine release. Additionally, LL-37 inhibits biofilm formation and stimulates angiogenesis and re-epithelialization in wound healing models.

Overview

LL-37 is a 37-amino acid peptide and the sole cathelicidin-derived antimicrobial peptide found in humans. It is produced by cleavage of the precursor protein hCAP-18 (human cationic antimicrobial protein 18) and is expressed in neutrophils, macrophages, epithelial cells, and other immune cell types. In preclinical research, LL-37 has demonstrated broad-spectrum antimicrobial activity against gram-positive and gram-negative bacteria, fungi, and enveloped viruses. Its mechanism involves direct membrane disruption of microbial cells as well as immunomodulatory signaling through formyl peptide receptor-like 1 (FPRL1) and other pattern recognition pathways. Beyond its antimicrobial properties, LL-37 has been studied for its role in wound healing, anti-biofilm activity, and modulation of innate and adaptive immune responses.
